Not looking good for him in a nice match up against Jax:

Santonio Holmes (knee, quadriceps) is listed as questionable for Week 2.
Coach Rex Ryan expressed concern with Holmes' availability, calling ILB David Harris (toe) a better bet to play against the Jaguars. It's worth noting that the Jets signed Patrick Turner off the practice squad earlier this week, further confirming their worry about Holmes. It looks like he's headed for a game-time decision in what would be an excellent matchup versus Jacksonville.
 
Like mud said in coachnorms' thread, it's all up to the doc:

Santonio Holmes (knee/quad, questionable) indicated that his availability for Week 2 will be determined by Jets doctors.
Holmes also revealed that he suffered the injury in practice. We're purely speculating, but it's fathomable that the Jets might hold Holmes out of the game if they believe they can whip Jacksonville with the run game. "We’ll see come Sunday. We’ll see what the doc says on Sunday," Holmes said. The good news is Jaguars-Jets is a 1PM ET game. We'll know his status early.
